The line alone isn't particularly funny, but it certainly stands out due to the way Sarah (Sarah Buehler) delivered it and the context in which it was used. It was her most notable line throughout her appearance in that one episode.

While Wil posted the footage for his entertainment, he didn't consider the ramifications of his actions. This was similar to when he played on Penny's insecurities in "The Wheaton Recurrence" until she broke up with Leonard so that he could beat their bowling team. Although Wil was one of the best guest stars on The Big Bang Theory, he often caused tension between the main characters just by something he said, even when it was funny.

After Sheldon gets fired in "The Luminous Fish Effect" and starts taking up odd activities to fill his time, Leonard calls Mary to intervene. While talking about her son, she tells Leonard he got his temper from his father and has her eyes, but his unique intellect was from Jesus.

There's no question that Mary loved her son, even though most times she couldn't understand him and could only explain his unusual intelligence as a miracle. The line is even referenced later in the show in "The Maternal Combustion," when she mentions how she prayed to have a son smarter than his father and saw a Jesus bobblehead in the next car nodding yes.

In all his appearances, it was clear that Zack wasn't as intelligent as the rest of the main crew, and quotes like these further cement that character trait. Most times, Leonard and Sheldon made fun of him, much to Penny's embarrassment, while Zack remained completely oblivious.
