In an interview with Collider, Kripke spoke about the potential for a Supernatural reboot, responding that he would be interested in seeing it. He said that he “love[s] that universe” and would be willing to return to it if “there was something out there that really surprised” him. Of course, I’d want to see it. Whether I’m a part of it depends on, could I find something fresh about it that I have never seen before? Obviously, I’ve told a lot of those stories, but if there was something out there that really surprised me, I love that universe , and I’d be interested in looking at that. It’s tricky to find what story in that universe hasn’t been told, but if someone can find one, I’m all in, baby.

Therefore, given how close the Supernatural creative mind and the stars are, this only raises the likelihood that the Winchesters could be involved again in a new set of episodes if they can find the right story.

Supernatural had over 300 episodes in the main series, so it makes sense why Kripke is skeptical as to how many more stories can be told in the universe. The creator himself left as the showrunner in 2010 after completing his five-season story plan for the show. Furthermore, the franchise tried expanding its lore with The Winchesters, which was cancelled after only a single season. With The Winchesters' cancellation, it seems somewhat unlikely that a Supernatural reboot would be able to happen successfully without both Ackles and Padalecki involved.

Supernatural was originally supposed to end with season 5, but The CW wanted to continue the show and did so, leading to another 10 seasons.

However, a new update may make the likelihood of a Supernatural reboot more possible. The CW recently announced Walker's cancellation after four seasons, which Padelecki led. This means that the actor will now have a more open schedule and could return to a major franchise like Supernatural with a greater likelihood. Even Padalecki recently revealed that he was open to a revival, albeit sharing a similar condition that the story would need to be right and contribute to the canon established in the show.

While it is unclear how much the original duo and Kripke have discussed a potential reboot, there have been conversations about their reunion in The Boys. After Kripke and Ackles expressed a desire for Padalecki to appear on the Prime Video show, the creator recently said there are more active offers for Padalecki to appear, saying “texts have been sent.” Therefore, given how close the Supernatural creative mind and the stars are, this only raises the likelihood that the Winchesters could be involved again if they can find the right story.
