The James Gunn's DC Universe happens to be Clark Kent's nemesis, Lex Luthor.
While Nicholas Hoult's Lex Luthor will be the main villain Corenswet's Superman faces in the DCU movie, he will be far from the only threat the hero takes on. That is why Gunn added the Justice Gang to the cast of the Superman movie, as there are multiple foes that require more heroes besides Superman to stop them. One surprising villain Superman faces in the movie is a kaiju that is attacking Metropolis. However, I don't think the monster poses a threat to his life. That said, there are four characters in the Superman movie trailers who could kill him.
Sun-Like Monster
The Superman Movie's Apparent Final Villain
So far, most Superman movie trailers build to a dark moment between the Man of Steel and Lois Lane. Corenswet's Clark Kent and Rachel Brosnahan's Lois seem to be saying goodbye before he flies out of the window of their apartment to face a mysterious threat on the horizon. At the moment, not much is known about the villain in question besides the fact that it looks like the sun and is seemingly quite powerful. Nathan Fillion's Guy Gardner is fighting the monster in the background of the somber scene with Lois and Clark.
Despite the Green Lantern's efforts to contain the creature with a net made out of his powers, that does not seem to be enough, requiring Superman to fly off to help. Judging by the way the trailers build towards that moment at the end, the pink energy the creature fires, and how the scene with Lois and Clark in their apartment is quite dark, I feel like the sun-like monster will be the movie's final villain, and thus Superman's strongest foe. The monster could be Solaris, an artificial and sentient sun that is key to All-Star Superman, which inspired Superman.
Ultraman
The Most Likely Main Physical Threat Of The Superman Movie
While the sun-like monster, which could be the DCU version of DC Comics' Solaris, looks to be the most powerful threat in James Gunn's Superman movie, I'm convinced that Ultraman will be the main physical threat that the Man of Steel fights throughout the film. Ultraman can be seen fighting David Corenswet's Superman in the DC movie's trailers, as well as arresting the hero alongside Rick Flag Sr. and the Engineer. As such, it will be interesting to see where his allegiances lie. Ultraman's secret identity has yet to be revealed.

A Character That Connects To An Impotant Superman Movie Plot Point
Like Ultraman, the identity of the Hammer of Boravia is also a secret at the moment. The character should play a decent role in Superman, as the Hammer of Boravia is connected directly to one of the film's main storylines. In the latest Superman movie trailer, it is revealed that David Corenswet's Superman intervened in a war in another country, leading to the world seeing his actions as a sign of the United States making a choice. That lands the Man of Steel in hot water, and the Hammer of Boravia is key to that storyline.

David Corenswet's Superman might look weak in the trailers for the DCU movie, but I believe it is the best way to move past Henry Cavill's Superman.
The fictional nations that go to war are Boravia and Jarhanpur. The Hammer of Boravia might be angry that Superman interfered when he should not have, leading to him going after the Man of Steel in Metropolis, as seen in the Superman movie trailers. What is interesting is how powerful the mysterious character is. At one point, the Hammer of Boravia uses heat vision, leading to theories about how characters like Supergirl, Bizarro, Doomsday, and even Ultraman could be behind the character's large armor. Judging by his sheer size and powers, the Hammer of Boravia will be hard to stop.
Lex Luthor
The Superman Movie's Main Villain Is The Hero's Classic Nemesis
Finally, Nicholas Hoult's Lex Luthor is another deadly threat to David Corenswet's Superman. While the Superman movie trailers do not show Lex actively fighting the Man of Steel, he is the DCU movie's main villain. As such, he will be pulling the strings behind the scenes and is likely responsible for multiple threats the hero will face. One of the deadliest of them is María Gabriela de Faría's Engineer. The character is seen transforming parts of her body into buzz saws and more with the help of nanotechnology, which Lex Luthor likely had a hand in.

The Engineer is not the only character working for Lex in the DCU's Superman movie. Anthony Carrigan's Metamorpho is an interesting player. Corenswet's Superman will be locked up in a high-tech cage at some point in the movie, and it seems like Metamorpho is using his powers to transmute into any known element to turn his hand into kryptonite. Given how there is a woman in the cell next to him, I'm convinced Lex Luthor is forcing Metamorpho to weaken Superman by threatening the woman, who could be Sapphire Stagg, Metamorpho's love in the comics.
As such, Lex Luthor can control a deadly fighter in the form of the Engineer and has access to Superman's biggest weakness, kryptonite, through Metamorpho. However, Hoult's DCU villain could be a major threat to the Man of Steel by himself, too, as new Superman movie merch revealed Lex Luthor's warsuit from DC Comics. It seems like the villain will take to the skies in a suit that allows him to trade blows with David Corenswet's DCU hero. With his genius-level intellect and a powerful suit of armor, Lex Luthor could be strong enough to kill Superman.
